If I am going to Costco to purchase stuff for my own family, but also purchase stuff for daycare (say I choose to go to Costco because I know I need items for both daycare and my family) is that mileage deductible?

You can only deduct a trip if the "primary" purpose of the trip is business. You must be able to say that at least 51% of the reason for the trip is business, or you must buy more business stuff than personal stuff.
